OPA/FMOC-protocol for AAA

The OPA/FMOC-protocol for AAA workflow allows you to perform the derivatization of the amino acids directly in the needle/loop prior to injection. This procedure is described in the Application Note 5994-2189.

The sample is taken from the position specified in the sample information or sequence/worklist. The mixture is prepared within the needle and directly injected. For more details, mark the Advanced check box at the top right of the workflow view to display the injector program in the advanced view.

Parameters

The following parameter settings are available:

Parameter	Description
Sample Volume	Specify the sample volume.
Borate Buffer Volume	Specify the volume of the borate buffer.
From Location	Enter the location of the borate buffer. See Specifying a Location.
OPA Reagent Volume	Specify the volume of the OPA reagent.
From Location	Enter the location of the OPA reagent. See Specifying a Location.
Mix 1 Volume	Specify the air volume for the mixing within the needle.
Repetitions	Select the number of repeat cycles for the mixing.

FMOC Reagent Volume	Specify the volume of the FMOC reagent.
From Location	Enter the location of the FMOC reagent. See Specifying a Location.
Mix 2 Volume	Specify the air volume for the second mixing within the needle.
Repetitions	Select the number of repeat cycles for the mixing.

Injection Diluent Volume	Specify the volume of the injection diluent.
From Location	Enter the location of the injection diluent. See Specifying a Location.
Mix 3 Volume	Specify the air volume for the third mixing within the needle.
Repetitions	Select the number of repeat cycles for the mixing.

Accumulated Draw Volume	The sum of all entered volumes plus 0.7 µL per standard wash cycle. The maximum draw volume is limited by the loop volume and the syringe volume. If the accumulated draw volume exceeds the maximum draw volume, a warning icon is displayed.
