Dilution Series

The Dilution Series workflow allows you to generate a dilution series from a concentrated sample before injection. You specify a dilution factor by which all dilution steps are calculated. The sampler pipettes then the series, starting from the specified sample location and dilution solvent location, into the first target location. Then it takes each dilution step from the previous dilution. The specified mixing method is used to mix the diluted samples in the target sample vial.

This workflow is similar to the Dilution Series from Stock workflow, but it takes each dilution step from the previous dilution. If you want to use a dilution series workflow that takes each dilution step from the sample stock solution, use the Dilution Series from Stock workflow.

Example

Draw Sample

The Draw Sample parameters specify the location and the injection volume of the sample.

Parameter	Value	Description
From Location	Sample	The sample is taken from the position specified in the sample information or sequence/worklist.
From Location	Location	Enter the location in the adjacent field. See Specifying a Location.
Sample Volume	Specify the volume to be drawn from the sample location.

Draw Dilution Solvent

The Draw Dilution Solvent parameters specify the dilution factor and solvent with which to dilute the sample.

Parameter	Description
From Location	Enter the location of the dilution solvent. See Specifying a Location.
Dilution Factor	Select the dilution factor.
Diluent Volume	The volume of the dilution solvent as specified in the Sample Volume and Dilution Factor parameters.

Dilute

The Dilute parameters specify the target and the mixing of the diluted sample.

Parameter	Value	Description
Target Location	Enter the target locations. See Specifying a Location. Click Add to add a location or Remove to remove the selected entry.
Sample Volume in Target	The resulting sample volume in the target.
Mix Mode	Select the mixing method that is used to mix the diluted sample in the target location.
	Air bubble	The air is taken from a neutral position (e.g. wash port) and bubbled into the diluted sample. This method should be the default, as it is the most effective. For some samples, especially those containing detergents or proteins, the method may cause the sample to foam. When the sample is drawn up later, the foam may cause insufficient sample volume to be drawn up. In this case, use the other method.
	Draw/inject from/to sample	The maximum volume of the diluted sample is drawn from and ejected in the target location.
Repetitions	Select the number of repeat cycles for the mixing method.
Speed	Specify the eject speed for the mixing with mix mode Draw/inject from/to sample.
	Default	The eject speed specified in the Set Up panel.
	Maximum	The maximum speed that the sampler can handle.
	<value> (µL/min)	A value that overrides the default eject speed.
