Learn more about method configurations
NOTE
Processing methods typically contain many time-based parameters, such as expected retention times or retention time windows. Such methods are designed for data that had been acquired with a certain flow, column dimension and other acquisition parameters.
If you want to apply such methods to other data with different acquisition parameters, you must adapt the method settings.
The selected method configuration determines which sections of the method are visible and can be edited in the Processing Method window.
2D-LC methods contain the following sections:
General:
Properties section: Same function as for 1D data.
Signals section:
In the Alignment tab, you adjust the delay of your ¹D and ²D detectors (see Align signals over two dimensions).
In the Blank Subtraction tab, you specify how the ¹D signals of blank runs will be subtracted from the ¹D signals of sample runs.
In the Blank/Cut Subtraction ²D tab, you specify how the ²D signals of blank runs will be subtracted from the ²D signals of sample runs.
The settings in the Smoothing and Reference Chromatogram tabs only apply to ¹D signals.
Extraction:
Under Extraction, you specify the parameters for:
MS and UV chromatogram extraction from ¹D data
MS Chromatogram extraction from ²D data (see Extract Chromatogram for MS data recorded in ²D)
MS and UV spectra extraction from ¹D data
MS and UV spectra extraction from ²D data (see Extract an MS spectrum from a ²D signal, and Extract a UV spectrum from a ²D signal)
Integration Events: Same function as for 1D data
2D Peaks:
Compounds:
Identification section, Compound Table tab: Same function as for 1D data, plus renamed and additional values.
Qualifier Setup tab: These settings apply only to ¹D signals.
General tab: Additional option to use ²D time reference compounds.
Calibration section:
Same function as for 1D data.
Spectra section:
These settings apply only to ¹D signals.
Spectra ²D:
System Suitability
Same calculations as for 1D data. However, there are a few differences in the workflow and output. See System Suitability and Noise calculation for ²D peaks.
Reports
Same function as for 1D data. Choose a 2D-LC report template.
Custom Calculation
Same function as for 1D data, plus 2D-LC fields and functions.
