Prepare for manual tuning (LC/MS)

Preparation for manual tuning consists of loading the atunes.tune file, making sure the Single Quadrupole LC/MS is set for the correct Ion Source and Polarity (Positive or Negative), and making sure the parameters that control ion formation are at acceptable settings. When tuning with a MultiMode source (MMI), tune in MMI-APCI mode.

  1. Click Method.

  1. In the Acquisition Method window in the navigation pane, click Instrument Setup > SQ.
  2. In the navigation pane, double-click Tune > Manual Tune.
  3. If needed, click Take. In this image, Take is already selected.

  1. In the navigation pane, select Tune > Manual Tune > Source.
  2. (optional) Open a different tune file. Click to open the Load Tune File dialog box. Select a tune file and click Load.
  3. Click to lock the instrument for tuning. You need to unlock the instrument after you finish tuning.
  4. If the source is MMI, then select the Ion mode. You can select ESI, APCI, or ESI+APCI.
  5. Turn the Calibrant on. Select the Calibrant on check box. Use the appropriate calibrant.
  1. Click the Polarity. You can click Positive or Negative.
  2. The Source Parameters change depending on which source is installed. The default source parameters are loaded for the installed source. These values are good starting values:
    Because of slight variations from spray chamber to spray chamber, changes such as adjustment of the nebulizer, and normal aging of LC/SQ components, these default values may not be optimum for a particular spray chamber. You can refer to your Acquisition method for other starting values.
