In the context of sample purification, users often perform fraction collection for analytical or preparative purposes. With fraction collection, compounds separated during the run by the chromatographic process are recovered into individual tubes.
Fraction collection can be performed on LC systems fitted out with a fraction collector module and one or several detectors (UV, DAD, mass detector (SQ), etc.)
When a mass detector is used to trigger the fraction collection, we are talking about mass-based fraction collection (MBFC). MBFC concerns LC instruments with a fraction collector and an MS detector. The system can also include an additional detector such as UV.
The following MBFC workflows are supported:
Use MBFC parameters defined in the acquisition method
MBFC dedicated parameter values are only defined in the acquisition method. If you want to collect fractions, for example, based on different target masses, define different acquisition methods in the sequence lines.
Override the method target masses and threshold values in the sequence
MS target masses and/or fraction collector thresholds values are defined in each individual sequence line. You can use the same acquisition method in all sequence lines, as the values in the acquisition method are overridden by the ones defined in the sequence.
