Oligo Analysis Accelerator (OAA) for OpenLab CDS is a software add-on to streamline the data analysis for the determination of purity and assay for synthetic oligonucleotides. A step-by-step wizard guides you to perform what otherwise would be laborious and intensive steps performed in native data acquired with an Agilent LC/MSD single quadrupole detector.
To prepare for the analysis, an OpenLab CDS processing method is applied to the data set. The software wizard then guides you through relevant steps.
To access the OAA web interface, open https://localhost:52088/openlab/oaa/login on a client where the Add-on is installed.
The overall workflow includes the following steps:
Perform a negative check out.
In Acquisition, prepare an acquisition method.
In Acquisition, set up a sequence.
Acquire data.
In Data Analysis, prepare result set and processing method.
In the OAA web interface, set up OAA project parameters.
In the OAA web interface, run the oligo analysis.
The oligo analysis includes the following steps:
Initial Setup: Apply project settings to the result set. Complete data analysis method parameters for system suitability and sample, and apply them as well.
System Suitability: Confirm UV integration for QC standards and adjust if necessary. UV and MS calibration curves are generated, and MS results are summarized. Check the pass/fail result for each SST parameter. Results are reported in a summary page.
Ion Classification: Ions that exceed the threshold are automatically classified as either known, unknown or adducts. Spectra from both standard and harsh MS sources are available if further investigation is needed. Confirm the classification and adjust if necessary. A final classified ion list is reported prior to extraction.
Integration: Extracted ion chromatograms from classified ion list are integrated. Also, UV signals for each sample are integrated. Adjust EIC and UV integration if necessary.
Summary: Check and confirm the summary of purity and assay calculations.
In Data Analysis, report the results. Apply eSignatures and approvals as required.
